摘要:
采用乙醚连续渗漉提取白秃马勃粗脂肪,利用硅胶柱色谱对白秃马勃粗脂肪化学成分进行分离纯化,通过气相色谱 - 质谱联用技术(GC-MS)分析菌油脂肪酸成分,运用电喷雾电离质谱(ESI-MS)、一维核磁共振谱(1DNMR)和二维核磁共振谱(2DNMR)解析化合物结构,通过体外实验测试菌油对 1,1- 二苯基 -2- 苦基肼基(DPPH) 的清除能力。 结果表明, 白秃马勃菌油主要脂肪酸为亚油酸(44.11%)、 油酸(27.49%) 和棕榈酸(17.60%)。 2 个单体化合物被解析为麦角甾 -5,8- 表二氧 -6,22- 二烯 -3- 乙酸酯(MB1)和麦角甾 -5,8- 表二氧 -6,22- 二烯 -3- 醇(MB2)。 白秃马勃菌油在试验浓度范围内具有一定的 DPPH 自由基清除能力,半数清除浓度(IC50)为 26.61mg/mL,并且呈浓度依赖关系。 化合物 MB1 和 MB2 均为首次从白秃马勃菌分离得到。
Abstract:
ThecrudefatofthepuffballCalvatia candida (Rostk.)Hollóswasextractedbyetherandpurifiedbysilica column chromatography. The fatty acid composition of the fungal oil was analyzed by gaschromatography-mass spectrometry (GC-MS), andthe compound’s structure was elucidated by electrosprayionizationmassspectrometry(ESI-MS),one-dimensionalnuclearmagneticresonancespectroscopy(1DNMR)andtwo-dimensionalnuclearmagneticresonancespectroscopy (2DNMR),andthescavengingactivityofthefungaloilagainst1,1-diphenyl-2-bitterhydrazine (DPPH)wastestedinvitro.Theresultsshowthatthemainfattyacidsinthefungaloilwerelinoleicacid (44.11%),oleicacid (27.49%)and palmiticacid (17.60%),respectively.Thetwomonomercompoundswereelucidatedasergosta-5,8-phenylenedio-6,22-diene-3-ylacetate (MB1)andergosta-5,8-phenylenedio-6,22-diene-3-ol (MB2),respectively.ThefungaloilhadDPPH scavenging activity in the range of experimental concentration with the half scavenging concentration(IC50) of 26.61 mg/mL, and showed concentration dependence. Both the compounds MB1 and MB2 wereisolatedfromC. candida forthefirsttime.
